abstract The present invention provides methods for preparing protein conjugates from ketone-modified proteins. In one embodiment, prepared by linking the sulfur atoms of two cysteines together via reaction with 1,3-dihaloacetone or similar ketone-containing reagents to link two free cysteines the protein. The ketone inserted between the sulfur atoms is then used to form an oxime, thereby conjugating the protein to the payload. In another embodiment, two cysteines are linked together by reaction with 1,3'-dihaloacetone or similar reagents, and the new ketone is used to form an oxime with a suitable payload molecule. This method provides improved reaction conditions for oxime formation, resulting in high yields and improved product uniformity.
