The recombinant Escherichia coli is obtained by dehydrating glycerol dehydratase gene dhaB123, glycerol dehydratase reactivation factor gene gdrA and α-ketoglutarate semialdehyde The hydrogenase mutant encoding gene TUkgsadh and the glycerol triphosphate dehydrogenase encoding gene gpd1 are introduced into the host bacteria and constructed. By using gene knockout technology, the inhibition of glycerol entering and leaving cells is relieved, and the cofactor NAD + of aldehyde dehydrogenase is regenerated at the same time. This results in a 12-fold increase in 3‑HP yield.
