abstract | The application discloses a joint processing method applied to Small RNA second-generation sequencing library building and application thereof. The joint processing method of the present application includes: A. adding a SE library 3 'joint and a 5' joint at two ends of a Small RNA fragment respectively; B. carrying out reverse transcription on the Small RNA fragments connected with the joint to obtain cDNA; C. adding "A" to the 3' end of the reverse transcribed cDNA fragment; D. and C, connecting a Y-shaped joint of PEI to the cDNA fragment of the step C. According to the connector processing method, the PEI connector is added on the basis of the SE connector, so that a PEI sequencing reagent can be used in sequencing of the constructed Small RNA library, the problem that the sequencing reagent of the SE library and the PEI library is incompatible is solved, sequencing of the Small RNA is facilitated, and convenience is provided for cancer diagnosis and subsequent targeted therapy based on the sequencing of the Small RNA. |
