When the target is UDG, if UDG exists, the U base in the hairpin probe is removed to generate an AP site, and the generated AP site is cut by the tool enzyme EndoIV to release the free 3' The primer sequence at the end is used to initiate the subsequent rolling circle amplification reaction RCA; if UDG does not exist, the 3' end is blocked in the hairpin probe, and the RCA process cannot proceed; when the target is EndoIV, if EndoIV exists, The RCA process is the same as the RCA process in the above-mentioned UDG activity detection. If EndoIV does not exist, the 3' end is blocked in the hairpin probe, and the RCA process cannot be performed. It can be used as both a recognition probe for UDG and a precursor recognition probe for EndoIV, thus simplifying probe design. The detection limits of UDG and EndoIV were as low as 0.00017 U/mL and 0.11 U/mL, respectively, and the results were better or comparable to other label-free fluorescence methods.
