abstract The present invention discloses a 3-indole carboxylic acid preparation method comprising the following steps: (1) indole and trichloroacetyl chloride are added into methylene chloride for reaction under the effect of anhydrous aluminium trichloride to obtain crude 3-trichloroacetyl indole; (2) the crude 3-trichloroacetyl indole is poured into ice water, stirred and allowed to stand for layering to separate an organic layer; (3) the organic layer is dried, filtered and concentrated to obtain pure 3-trichloroacetyl indole; (4) the 3-trichloroacetyl indole is added into methanol, an alkali solution is added dropwise for heating for refluxing for 18 to 22 hours, the mixture is cooled to room temperature, the methanol is recovered by concentration, and a remaining liquid is remained; (5) water is added into the remaining liquid, hydrochloric acid is added dropwise for adjusting PH to 3-4, the remaining liquid is filtered, and a solid obtained by the filteration is dried to obtain crude 3-indolyl carboxylic acid; and (6) the crude3-indole carboxylic acid is beaten with ethyl acetate, then filtered, and dried to obtain pure 3-indole carboxylic acid. The 3-indole carboxylic acid preparation method provides a new idea for the preparation of 3-indole carboxylic acid, and significantly improves product yield and purity.
