The preservation number of this engineering bacterium is CGMCC No.11460, and the classification name is Escherichia coli. The invention mainly discloses a thermophilic alkaline pectin lyase, the optimum reaction temperature is 70°C, the optimum Ca2 + concentration is 0.8mM, the substrate is 0.2% (w/v) polygalacturonic acid, pH 9.5 . The specific activity of the enzyme is 900U/mg at optimum pH 9.5, Ca 2+ 0.8mM, and 70°C; at 70°C, the half-life is 6h; after purification by nickel column, CbPelC can reach 50mg/L (1L fermentation liquid). The heat resistance and yield of the thermophilic alkaline pectin lyase in the invention are significantly higher, more suitable for modern industrial production requirements, and also provide an effective method for large-scale industrial production of alkaline pectinase.
