abstract The invention provides a method and kit for determining mutated nucleic acid bases and relates to the technical field of nucleic acid amplification. The method and kit are mainly used for more accurately and sensitively determining whether mutated nucleotide exists in an authentication area of a target nucleic acid sequence in a sample. The method comprises the step a of providing a first primer, a second primer and an oligonucleotide probe which contains a radical capable of being detected and has a mark of a sequence based on an amplification area of the target nucleic acid sequence, wherein the first primer can be located in the authentication area of the target nucleic acid sequence containing the mutated nucleotide in a hybrid mode; the step b of performing an amplification reaction through a PCR; the step c of determining an amplification product through an amplification curve; if the target nucleic acid sequence contains mutated nucleotide, the first primer performs hybrid with the target nucleic acid sequence containing the mutated nucleotide, and then an amplification reaction can be generated; if the target nucleic acid sequence does not contain mutated nucleotide, the first primer performs hybrid with a normal target nucleic acid sequence, and then an amplification reaction cannot be generated.
