abstract Disclosed are unnatural amino acids and polypeptides comprising at least one unnatural amino acid, as well as methods of making the unnatural amino acids and polypeptides. The unnatural amino acid itself or as part of a polypeptide can include a wide variety of possible functional groups, but typically has at least one aromatic amine group. The invention further discloses post-translationally modified non-natural amino acid polypeptides, methods for realizing the modifications and methods for purifying the polypeptides. Such modified non-natural amino acid polypeptides typically include at least one alkylated amine group. In addition, the invention discloses methods of using the non-natural amino acid polypeptides and modified non-natural amino acid polypeptides, including therapeutic, diagnostic and other biotechnological uses.
