titleOfInvention | Soluble pyrroloquinoline quinone dependent glucose dehydrogenase mutant with improved glucose specificity, polynucleotide and application thereof |
abstract | The invention provides a soluble pyrroloquinoline quinone dependent glucose dehydrogenase (PQQ-sGDH) mutant, the amino acid position of which corresponds to the wild-type PQQ-sGDH sequence of Acinetobacter calcoaceticus SEQ ID NO1, wherein the PQQ-sGDH mutant carries a mutation comprising one of the following group of mutations: T372C, T372N, a combination mutation based on T372C and a combination mutation based on T372D. These PQQ-gdh mutants have good substrate specificity for glucose and significantly reduced cross-reactivity with sugars such as maltose, and are particularly suitable for the detection of glucose in samples such as blood. |
