abstract | The invention discloses a strain of recombinant Escherichia coli, which is named as Escherichia coli (Escherichia coli) pCodonPlus/pRKISC/pETDuet‑S33[2Fe2S]Fd, which contains [2Fe2S]Fd derived from Agrobacterium tumefaciens S33 ] ferredoxin gene, the nucleotide sequence of which is shown in SEQ ID NO.1; the recombinant Escherichia coli is to connect the [2Fe2S] ferredoxin gene into the pETDuet-1 vector, and the recombinant plasmid is named pETDuet ‑[2Fe2S], and transfer the obtained recombinant plasmid into E.coli C41 (DE3) containing pCodonPlus and pRKISC plasmids. The present invention also discloses the application of the recombinant Escherichia coli in the fermentative production of [2Fe2S] ferredoxin. Experiments have confirmed that 23 mg of purified [2Fe2S] ferredoxin can be finally obtained per liter of fermentation culture, which is much larger than before The reported yield of 0.03 mg purified from wild bacteria indicates that the recombinant Escherichia coli provided by the present invention has a good application prospect in industrial production. |
