abstract | The invention provides an ADP-glucose pyrophosphorylase mutant and a screening method and application thereof, wherein the screening method comprises the following steps: 1) synthesizing first strand cDNA; 2) PCR amplification; 3) constructing an AGPase subunit cDNA prokaryotic expression vector; 4) and (4) activity screening. The invention carries out distant hybridization on AGPase large subunit genes and small subunit genes from different species, and obtains an expression vector of a recombinant gene with high controllability; then transforming the escherichia coli glgC mutant strain, inoculating the strain on a Conberg enriched solid culture medium, and finally, using iodine-potassium iodide to dye and screen out the high-activity ADP-glucose pyrophosphorylase mutant; the ADP-glucose pyrophosphorylase mutant has enzyme activity and glycogen content obviously higher than those of wild corn AGPase, does not obviously change substrate affinity, and increases sensitivity to an activator. |
