abstract The invention discloses a kind of α ptyalases enzyme detection device based on potentiometry and prepare application method, and it is used successfully to detect the alpha amylase concentration in human saliva, principle is that starch decomposes generation maltose in the presence of α ptyalins, and redox reaction occurs in the basic conditions for the maltose and the potassium ferricyanide with reproducibility:Maltose+the potassium ferricyanide → maltobionic acid+potassium ferrocyanide.Electrode potential is determined by the concentration ratio of the potassium ferricyanide and potassium ferrocyanide, therefore the potassium ferrocyanide of generation can cause the potential change of respective electrode, i.e. alpha amylase activity is higher, measures that potential change is bigger, and the activity of α ptyalins can be calculated by measuring potential change.Relative to traditional salivary alpha amylase method, fast and convenient using potentiometry detection, required reagent stability is good, and the electrochemical detection system preparation technology of two electrodes composition is simple, and required detection circuit is simple, and low in energy consumption, cost is low, has good application value.
