abstract | The present invention relates to the controlled release of substances from porous substrates such as mesoporous silica nanoparticles. Particles comprise guest molecules (eg pharmaceutical agents or contrast agents) trapped within the pores of the substrate. The pore is capped by at least one nucleic acid sequence and a reagent is released by a triggered reaction with the capping sequence with at least one analyte (biomarker) thereby allowing the cap to be cleaved from the pore. The capping agent may be a DNAzyme, which cleaves its substrate and releases the guest molecule upon binding to the analyte. Alternatively, the capping agent may be a hairpin oligonucleotide, which is digested by a nuclease after hybridization to a complementary nucleic acid analyte. The invention also relates to methods for the manufacture of said substrates, their use for the controlled administration of active agents and the diagnosis of conditions in patients. |
