abstract | A method for multiplex ligation-dependent probe amplification comprising (a) providing a sample tissue containing different target nucleic acids, (b) providing a different probe set for each target nucleic acid, each probe set comprising a sequence having a first adaptor and a first locus-specific probe of a first target-specific moiety and a second locus-specific probe having a second linker sequence and a second target-specific moiety adjacent to said first target-specific moiety, ( c) hybridizing the probe set to the target sequence to form a hybrid complex; (d) ligating the hybrid complex to form a ligated probe, (e) amplifying the ligated probe to form an amplicon , and (f) detecting the amplicons in the detection system by sequencing each amplicon. |
