abstract | The present disclosure provides chips, systems and methods for sequencing nucleic acid samples. Labeled nucleotides are provided to the reaction chamber containing the nanopores in the membrane. A single labeled nucleotide of the labeled nucleotides may contain a label coupled to the nucleotide that is detectable by means of the nanopore. Next, a single labeled nucleotide of the labeled nucleotides can be incorporated into a growing strand complementary to a single-stranded nucleic acid molecule derived from the nucleic acid sample. By means of the nanopore, a label associated with a single labeled nucleotide can be detected upon incorporation of the single labeled nucleotide. The label can be detected by means of the nanopore when the label is released from the nucleotide. |
