abstract The present invention relates to and the foundation of a series of artificial fluorescent's element enzyme based on the artificial amino acid sequence that extracts, provide a kind of characteristics of luminescence by the comparison by the amino acid similarity based on the luciferase sequence from oar pin class in database as high brightness and stability of photoluminescence is high, the spectrum that is migrated to long wavelength side.In the present invention, the luciferase sequence from oar pin class in database is compared based on amino acid similarity, it is extracted centered on the amino acid high by the frequency on each position in corresponding sequence, artificial amino acid sequence is built, foundes a series of artificial luciferase group (ALuc).At this point, the first half and latter half of enzymatic activity region that are present in amino acid sequence are also compared by similitude together, the similitude of the amino acid in the two also improves.Brightness and the stability of photoluminescence of each enzyme of obtained luciferase (ALuc) group are evaluated, the amino acid sequence of the good ALuc of result is fed back and builds amino acid sequence again, thus carrys out the higher luciferase of synthesis performance (ALuc) group.These ALuc classes have luminosity high brightness and stability of photoluminescence raising or luminescent spectrum to long wavelength side migrate etc. the unprecedented excellent characteristics of luminescence.Also, by using artificial fluorescent's element enzyme (ALuc) class of the present invention, it is capable of providing the unprecedented excellent bioassay system in bioluminescent probe, double cross detection, luminous capsule of function of measuring etc. is improved.
