abstract | The present invention provides the antibody and its antigen-binding fragment exclusively combined with former convertase subtilopeptidase A/kexin 9 (PCSK9) with bigger affinity in acid pH in neutral pH ratio.The antibody of the present invention can have one or more amino acid changes compared to the antibody without pH dependence binding characteristics.For example, the present invention is included in the anti-PCSK9 antibody that one or more complementary determining regions have the substitution of one or more histidines.The antibody with pH dependence binding properties of the present invention, compared to the anti-PCSK9 antibody without pH dependence binding properties, the cycle neutralization that extension is temporally retained in animal target shows cholesterol-lowering activity.The antibody of the present invention therefore can be used for treating and cholesterol-associated raised HDL disease and illness, wherein the antibody of the present invention can be applied to sufferer compared to the antibody without pH dependence binding properties with relatively low-dose and/or lower frequency. |
