abstract One aspect of the invention generally relates to directing selection, combination and expression of antibody variable domains entirely in vivo. One application is the production of multivalent polypeptides. The invention relates to multivalent (eg, multispecific) antibodies, antibody chains and polypeptides, as well as heavy chain-only antibodies lacking light chains (H2 antibodies). The invention further relates to the selection, maturation and production of these antibodies in non-human vertebrates and in non-human vertebrate cells. For this reason, the invention also relates to such non-human vertebrates and cells. The present invention also relates to providing means for generating and selecting heavy chain-only antibodies and heavy chains comprising variable domains that have undergone affinity maturation.
