abstract | The invention provides a novel porcine-derived antimicrobial peptide cecropin P1 mutant, its preparation method and application. The present invention uses biological software to analyze the spatial structure of the amino acid sequence of the porcine antimicrobial peptide cecropin P1, mutates two of its 31 amino acids (R10L, K12N), and adds asparagine at the C-terminal to facilitate the C-terminal Amidation of a new porcine-derived antimicrobial peptide mutant was obtained, and its antibacterial efficacy was significantly improved. On this basis, the new porcine antimicrobial peptide cecropin P1 mutant gene was artificially synthesized by using the preferred codons of Pichia pastoris, cloned and expressed in Pichia pastoris, and the recombinant yeast strain of the new antimicrobial peptide was obtained, and the fermentation scale was enlarged to a fermenter Level, to achieve high-density fermentation and high-efficiency expression of antimicrobial peptide products. After further purification, the fermentation broth can be made into powder, liquid and other antimicrobial peptide preparations for the prevention and treatment of livestock and poultry diseases. |
