abstract | The invention relates to a method for preparing a monoclonal antibody for treating psoriasis. The method is characterized by comprising the following steps: analyzing cell factors which are specifically expressed in psoriasis cells by using an isotope labeling and multi-dimensional liquid chromatogram-mass spectrum combination method, and separating and purifying the protein; immunizing a female BALB/c mouse of 6-8 week old by using the separated and purified cell factors which are specifically expressed in the psoriasis cells; if the antibody titer is greater than 1:10000, combining and screening a single-cell suspension of immune mice splenocyte and myeloma cells in the logarithmic phase, thereby obtaining hybridomas of a monoclonal antibody for secreting the cell factors specifically expressed in psoriasis cells; and injecting the hybridomas into the abdominal cavity of the BALB/c mice of 6-8 week old, collecting the ascites within about 10 days after the hybridomas are injected, and purifying the ascites by using a protein G/A column, thereby obtaining the relatively high-purity monoclonal antibody for treating psoriasis cell factors. |
