Processes for the preparation of treprostinil and derivatives thereof are described. Compared to existing techniques, this method utilizes the easily scalable enzymatic resolution of key intermediates used to prepare these compounds. Another significant improvement of the described method over existing methods is the regioselective Claisen rearrangement of the 5-allyloxy-benzaldehyde precursor, which is facilitated by the bromine substituent at the 2-position. |
