abstract | The method discloses a method for preparing high-yield leucine aminopeptidase through the fermentation of recombinant bacillus subtilis, which belongs to the fields of enzymic preparations and fermentation technologies. According to the invention, a culture method suitable for recombinant bacillus subtilis is obtained through combined optimization of culture medium optimization and fermentation process control; after being inoculated to a seed culture medium, the recombinant bacillus subtilis is subjected to shake cultivation until the logarithm growth period is reached, a certain amount of seed solution is inoculated to an adjusted fermentation culture medium, then the seed solution is subjected to fermentation cultivation by using a fermentation tank, and an operation of sampling is performed every 3h so as to determine parameters such as enzyme activity. According to the invention, a strategy of combination optimization is adopted for optimizing the conditions of producing leucine aminopeptidase through the fermentation of recombinant bacillus subtilis, then a fermentation culture medium and fermentation culture conditions are combined for optimizing, so that the ability of producing leucine aminopeptidase through the fermentation of recombinant bacillus subtilis is greatly improved, and the enzyme activity of leucine aminopeptidase is 605 U/mL. |
