abstract | The invention discloses a preparation method for expressing recombinant human growth hormone (rhHGH) in Escherichia coli in an inclusion body form. The method mainly comprises the following steps: carrying out high pressure homogenate crushing on thallus cells, rinsing the inclusion body, carrying out denaturation dissolving on the inclusion body by using urea, carrying out speed control grading renaturation on inclusion body proteins, centrifuging, carrying out ultrafilter membrane concentration, carrying out filtering bacterium removal, and freeze-drying to obtain a finished product. The recombinant human growth hormone prepared through the preparation method has the advantages of high purity and high activity. The recombinant human growth hormone with high purity and high activity can be prepared through renaturation and urea denaturation by using speed control grading renaturation of the proteins, and an ultrafilter membrane technology. The preparation method can omit the chromatography step, substantially shortens man hour, and reduces the production cost. Pure rhHGH can be efficiently, simply and cheaply obtained in the invention. |
