The method comprises the specific process: firstly, carrying out separation and purification of the crude extract of huperzia serrata by adopting an alkaline alumina column with the filler particle size of 100-200 mesh or 200-300 mesh, carrying out dry column packing with the diameter-height ratio of 1:8-1:30, carrying out dry sample loading with the sample loading ratio of 1:10-100, carrying out elution with chloroform-methanol (80:1-8:1), merging the flowing parts containing huperzine A, and packing a silica gel column to continue to separate. Silica gel column process parameters comprise: with the filler particle size of 100-200 mesh or 200-300 mesh, carrying out wet column packing with the diameter-height ratio of 1:8-1:30, carrying out wet sample loading with the sample loading ratio of 1:10-100, and carrying out elution with the chloroform-methanol (80:1-8:1). The obtained product is subjected to acetone recrystallization to obtain the finished product. The process route is economical and practical, and is convenient to industrialize.
