abstract | The present invention provides compositions comprising phosphorus-containing tricyclic compounds, including phthalazin-1(2H)-one derivatives. This compound is a potent inhibitor of poly(ADP-ribose) polymerase (PARP), especially PARP-1 and potentially PARP-2, which also shows good cellular activity in inhibiting poly(ADP-ribose) oligomer formation. These compounds are useful as monotherapy or in combination with other therapeutic agents in the treatment of PARP-associated diseases such as cancer, inflammatory diseases and ischemic diseases. Accordingly, the present invention also provides a method of treating a PARP-associated disease comprising administering to an individual in need thereof an effective amount of a compound of the present invention. |
