abstract | The present invention discloses a method for selecting cells with zero fucose levels for use as host cells for expressing recombinant proteins. The method comprises: (d) exposing CHO cells to methotrexate (MTX) to introduce a genetic mutation in the cell population of CHO cells; (e) exposing the CHO cell population containing the mutant cells to a non-toxic fucose binding agent for a certain amount of time , binding the fucose binding agent to the fucose moiety of the cell membrane of the cell population, wherein the certain amount of time does not result in killing the cells; and (f) removing the fucose binding agent from the cell population containing the mutant cells Subpopulations of cells of the agent were selected to be used as host cells for expressing the recombinant protein, and the selected cells had zero fucose content. The present invention also discloses cells and cell lines useful as host cells for expressing recombinant proteins. |
