titleOfInvention Method for extracting cyanobacteria toxin from urban eutrophic cyanobacterial bloom water
abstract The invention discloses a method for extracting cyanobacteria toxin from urban eutrophic cyanobacterial bloom water. The method comprises the following steps: (1) adding methyl alcohol and glacial acetic acid into to-be-extracted water to ensure that the final volume content of the methyl alcohol is 40% and the final volume content of the glacial acetic acid is 3-5%, and carrying out continuous ultrasonic treatment at the frequency of 300 hertz for 15-20 minutes; (2) regulating the pH (potential of Hydrogen) of the water sample subjected to the ultrasonic treatment of the step (1) to 3, filtering by using a filter membrane with the pore size of 0.45 micrometers, centrifuging the filtrate in the weight of 6500g for 5-10 minutes and taking a supernatant; (3) carrying out solid phase extraction on the supernatant obtained in the step (2) by using a solid phase extraction column, leaching by using a methyl alcohol aqueous solution with the volume content of 40%, and eluting by using a methyl alcohol aqueous solution with the volume content of 95% to obtain eluent containing the cyanobacteria toxin; (4) obtaining the cyanobacteria toxin from the eluent obtained in the step (3). According to the method disclosed by the invention, by adopting a solid phase extraction technique, two main isomers of the cyanobacteria toxin in urban lake water, namely MC-LR and MC-RR, are continuously enriched, purified and extracted; the method has the characteristics of high product purity, easiness for operation, low cost and the like.
