abstract | The invention discloses a P3/X63-Ag8 (X63) hybrid tumor cell of a Balb/c mouse. The P3/X63-Ag8 (X63) hybrid tumor cell is characterized by being capable of generating a mouse anti-human monoclonal antibody (mAnti-HCRCN81) for human HCRCN81 protein. The monoclonal antibody can identify the HCRCN81 natural protein in a human cancer cell and a cancer cell in a human cancer tissue. A preparation method of the monoclonal antibody is characterized by sequentially comprising the following steps of (1) killing human HCRCN81 protein immunized Balb/c mouse, taking the spleen of the mouse, and preparing a spleen cell suspension; (2) carrying out cell fusion on the homologous P3/X63-Ag8 (X63) cell and the mouse spleen cell to prepare the P3/X63-Ag8 (X63) hybrid tumor cell, and screening and purifying the positive P3/X63-Ag8 (X63) hybrid tumor cell; and (3) culturing the positive P3/X63-Ag8 (X63) hybrid tumor cell in the step (2) by using an in vitro culture method to generate the mouse anti-human monoclonal antibody (mAnti-HCRCN81). The natural human HCRCN81 protein is characterized by the specific high expression in the cancer cells in the human cancer tissue, can be applied to the early diagnosis of the cancer, and therefore can be applied to the diagnosis of the cancer, and has the potential to be used as the target for the early diagnosis and treatment of the cancer. |
