abstract | The invention relates to a general method for extracting the total microbial genome form traditional vinegar fermentation process samples, and belongs to the technical filed of molecular biology. A technical issue to be solved by the invention is to provide a method for extracting the total microbial genome from traditional vinegar samples in different states comprising solid state fermentation, semi-solid state fermentation and liquid state fermentation. The method comprises steps of pretreating the vinegar fermentation samples, extracting microbial genome DNA, removing impurities, rinsing and concentrating. The method for extracting relates to in-situ extraction. The sample amount needed is less than 5 g. Addition of preparations such as protease, cellulase, helicase, etc. is not needed. The genome extracted has high purity and high quality, and can be directly used for subsequent molecular fingerprint analysis, metagenomics analysis, PCR (polymerase chain reaction) amplification of targeted genes and clone library construction. |
