abstract The invention discloses a method for synthesizing an acid-orange II artificial antigen. The method is characterized by comprising the following steps of: weighing acid-orange II, adding acetone to dissolve the acid-orange II, adding succinic anhydride and anhydrous pyridine, and heating for refluxing for 24 hours; (2) removing the acetone by rotary evaporation, drying to obtain a layer of substance in orange color at the bottom of a distillation flask, adding DMF (dimethyl formamide) to dissolve, adding NHS (N-hydroxysuccinimide) and DCC (dicyclohexylcarbodiimide), stirring by a magnetic stirrer at room temperature to obtain a reaction liquid, centrifuging, and removing a precipitate to prepare an acid-orange II activating intermediate product; (3) weighing BSA (Bovine serum albumin) or OVA (ovalbumin), dissolving the BSA or the OVA in PBS (poly butylene succinate), dropwise adding the acid-orange II activating intermediate product into a BSA-PBS solution or an OVA-PBS solution while stirring by the magnetic stirrer, and stirring overnight; (4) adding a reaction liquid obtained by stirring overnight into a dialysis bag, and dialyzing for 72 hours in the PBS in a refrigerator, wherein the dialysate is changed every 8 hours; and (5) centrifuging the reaction liquid in the dialysis bag after finishing dialyzing, and removing a precipitate to obtain the acid-orange II artificial antigen.
