The method comprises the following steps of 1, marking reservation time of standard particles having different particle sizes by the asymmetrical vertical flow-field fractionation technology and drawing a linear standard curve, 2, carrying out characterization of virus-like particles in different solution systems or under different reaction conditions by the asymmetrical vertical flow-field fractionation technology to obtain reservation time of the virus-like particles and virus-like particle aggregates, 3, calculating hydraulic radiuses of the virus-like particles and the virus-like particle aggregates by the linear standard curve, and 4, carrying out structure characterization according to the calculation result of the hydraulic radiuses of the virus-like particles and the virus-like particle aggregates. The method for characterization is stable, has good reappearance and is suitable for structure characterization of different virus-like particles and virus-like particle aggregates.
