abstract | The invention discloses a method for fast detecting concentration of Escherichia coli in a water body by adopting a high-performance liquid chromatography method. The method comprises the following steps of: 1) obtaining a plurality of Escherichia coli solutions with different known concentrations; 2) determining the compliance relation of response values between Escherichia coli and 2,5-dihydroxy-benzoic acid on a high-performance liquid chromatography instrument as a linear relation according to the following chromatography conditions, i.e., a chromatographic column is a C18 chromatographic column, a flowing phase comprises liquid A and liquid B according to the volume ratio of (10-20):(90-80), wherein liquid A is acetonitrile, the liquid B is a phosphoric acid water solution with volume percentage of 0.03-0.075; conducting isocratic elution; detecting the wavelength of 205-235nm; and 3) introducing a water body sample to be detected, and reading the concentration of Escherichia coli in the water body to be detected according to responded concentration value of the 2,5-dihydroxy-benzoic acid through the known linear relation curve. The method is simple to operate, and the detection only lasts for three hours; and the detection limit can achieve 10,000cfu/mL. |
