abstract The invention discloses a method for promoting colorectal cancer (CRC) distant metastasis by microRNA-29a through KLF4. The method comprises the following steps of 1, preparing a tissue sample, 2, carrying out RNA extraction and real-time quantitative reverse transcription PCR, 3, carrying out cell culture and transfection, 4, carrying out a luciferase test, 5, carrying out western blotting, 6, carrying out cell invasion and 7, carrying out statistical analysis. Expression of miR-29a in CRC tissue is related with CRC distant metastasis; miR-29a expression improvement points out poor prognosis and combination of miR-29a expression improvement and KLF4 expression decreasing further points out poor prognosis; and a mechanism research shows that miR-29a promotes CRC cell invasion by acting on KLF4. The method provides a theoretical basis for miR-29a-based CRC patient prognosis evaluation and for miR-29a inhibitor-based CRC distant metastasis treatment.
