abstract The invention discloses a large-scale preparing and purifying method of bacteria O-specific polysaccharide. The large-scale preparing and purifying method comprises the following steps of: (1) hydrolyzing of lipopolysaccharide into bacteria O-specific polysaccharide raw sugar; (2) the bacteria O-specific polysaccharide raw sugar purifying: first-time ultra-filtration, alcohol fractional precipitation, the second-time ultra-filtration and anion exchange; the anion exchange: adding a buffer solution to an ultra-filtration concentrated solution which is obtained by the second-time ultra-filtration, sampling to an anion exchange column, and using the buffer solution to leach the anion exchange column until ultraviolet absorption value is not lowered; using eluting liquid to elute, and collecting the eluting peak, i.e., a target solution; and (3) concentrating and freezing the target solution to obtain the purified bacteria O-specific polysaccharide. The large-scale preparing and purifying method of the bacteria O-specific polysaccharide disclosed by the invention is convenient to operate, low in preparation cost, larger in treatment capacity, suitable for large-scale and industrial production. Moreover, the product obtained by production is low in impurity content, and high in purity, so that the requirements of furthermore developing human-use vaccines are achieved.
