abstract The invention discloses a preparation method of Gentialutine, and the preparation method is simple and convenient to operate and low in pollution. The method comprises the following processing steps of: (1) smashing a Gentiana tibetica root raw material, soaking the smashed raw material in ammonium hydroxide, drying, then, extracting in a supercritical CO2 extraction pot, and analyzing the extract, wherein the CO2 flux is 1-3ml/min/g raw material, acetone is used as an entrainer, the flux is 0.2-0.6ml/min/g raw material, the extraction temperature is 30-60 DEG C, the pressure is 15-25Mpa and the extraction time is 1-4 hours; (2) dissolving the extract in an aqueous solution of hydrochloric acid, filtering, performing multi-stage molecule cut off by a nanofiltration membrane, extracting the obtained liquid by chloroform, and concentrating the crude extract; (3) mixing chloroform, methyl alcohol and triethylamine solution (ph7.3) which are used as a solvent system at a ratio of (3-8:(2-5):(3-10), taking the upper phase as a stationary phase and the bottom phase as a mobile phase, separating the crude extract by virtue of high-speed counter-current chromatography, collecting fractions according to the color spectrum, reducing pressure, concentrating and drying to get the Gentialutine. The Gentialutine prepared by the preparation method is high in product purity and easy for industrial production.
