titleOfInvention Clone of anthurium MYB transcription factor AN2-like gene and expression vector construction
abstract The invention discloses an anthurium MYB transcription factor AN2-like gene and plant expression vector construction for the same. The plant expression vector construction comprises the following steps of: extracting the total RNA (ribonucleic acid) of anthurium 'alabama', and performing reverse transcription to synthesise cDNA (complementary deoxyribonucleic acid); taking the cDNA as a template for designing a specific primer; obtaining the full-length cDNA of the AN2 gene by means of PCR (polymerase chain reaction), and connecting the full-length cDNA with pMD-18T carrier; and transforming the competent cell of escherichia coli DH5 alpha, and selecting a positive clone to obtain the anthurium AN2 gene, wherein the full length of the cDNA of the gene is 1237 bp, and the cDNA comprises an upstream sequence in front of an initiation codon and a poly-A tail; and the cDNA is connected to the plant expression vector Cam35S-gfp to construct a novel plant expression vector which is named as Cam35S-an2, and the gene is related to synthesis regulation for anthocyanin. Therefore, obtaining for the AN2 gene lays the foundation for transgenosis culture for the different spathe colour varieties of anthurium, and has important theoretical and practical significance.
