titleOfInvention Method for continuously preparing beta-cyclodextrin by adopting manila immobilized cells
abstract The invention discloses a method for continuously preparing beta-cyclodextrin by adopting manila immobilized cells, belonging to the technical field of production of beta-cyclodextrin. The method disclosed by the invention comprises the following steps of: weaving manilas into a manila curtain and subsequently treating the manila curtain, fixing cyclodextrin glycosyl transferase CGTase producing bacterium ATCC21783 on the manila curtain, shaking a flask and producing CGTase, taking the manila curtain out every 2-3 days, washing the manila curtain by using normal saline to remove aged and degraded colonies from the manila curtain and putting the manila curtain into a fresh culture medium for continuous culture; using the CGTase produced by fermenting in production of the beta-cyclodextrin according to industrial steps. The continuous production of CGTase is facilitated by the manila immobilized cells; and further the continuous production of beta-cyclodextrin is realized. The process can last for more than one month. According to the method disclosed by the invention, the manila serving as a material which is natural and easily available is used as an immobilized substrate for the first time and used for immobilized CGTase producing bacteria; and the method disclosed by the invention is simple in immobilizing process and prominent in continuous production effect. Compared with the existed preparation process of beta-cyclodextrin, the method disclosed by the invention has the advantages of shortening the enzyme production period of the strain, simplifying the preparation process of the beta-cyclodextrin and improving the production efficiency.
