abstract | The invention discloses a transgenic chlorella with high biomass and/or high lutein yield and a preparation method thereof. The invention provides a method for culturing the transgenic chlorella. The method is carried out by importing a vitreoscilla hemoglobin coding gene to a target chlorella to acquire the transgenic chlorella, wherein the biomass and/or lutein yield of the transgenic chlorella are/is greater than that of the target chlorella; and the amino acid sequence of the vitreoscilla hemoglobin is a sequence 2 in a sequence table. The experiment of the invention proves that a vitreoscilla hemoglobin coding gene expression cassette is imported to a wild chlorella through the method disclosed by the invention so that the transgenic chlorella is acquired; compared with the biomass of the wild chlorella, the biomass of the transgenic chlorella disclosed by the invention is observably improved by 38.81% in comparison with the lutein content of the wild chlorella; the lutein content of the transgenic chlorella is slightly improved by 5.93% in comparison with the lutein content of the wild chlorella, and the lutein yield of the transgenic chlorella is greatly improved by 47.19% in comparison with the lutein yield of the wild chlorella, so that the transgenic chlorella is a chlorella with high biomass and/or high lutein yield. |
