abstract | A marking method of applying an SSAP molecular marker technology to a peony comprises the following steps: designing primers, extracting peony genome DNA, performing enzyme digesting for peony genome DNA, preparing a connector, connecting enzyme digested product with the connector, pre-amplifying of connected product, conducting a selective amplified reaction, testing polyacrylamide gel electrophoresis and the like. According to the marking method, pony genus retrotransposon LTR sequence primer is designed, and through the test of the polyacrylamide gel electrophoresis, stripes are rich; and compared with LTR primer of other species, the primer can directly used to polymorphism analysis between pony genuses, so as to achieve high efficiency and practicability; the selective amplified system is optimized and more suitable for the amplification of pony genus SSAP molecular marker; and silver staining method of polyacrylamide gel electrophoresis is improved, and fixing and dying steps of silver staining are combined to further improve the efficiency of applying the SSAP molecular marker on the pony genus. |
