abstract | The invention discloses a streptavidin mutant and a preparation method thereof. The nucleotide sequence of the streptavidin mutant is represented by SEQ ID No.6 in the sequence table. The amino acid sequence of the mutant is represented by SEQ ID No.7 in the sequence table. The mutant comprises mutations of 5 amino acids, which are E14D, T42Y, Y43T, S52G, and R53D. The invention provides the streptavidin mutant and the preparation method thereof. With the streptavidin mutant SA122m5, the affinity with biotin derivatives such as 2-iminobiotin is improved, and biotinylated product detection is facilitated. Half-life period of the mutant combined with biotin is prolonged, such that experiments and image researches requiring long-time observation are facilitated. |
