abstract Insufficiency of the proteasome to efficiently eliminate harmful proteins and prevent the formation of disease-causing aggregates leads to proteinopathies. As described herein, inhibition of the proteasome-associated deubiquitinase Uspl4 can enhance proteasome potency. Accordingly, the present invention provides novel compositions and methods for inhibiting Uspl4, enhancing proteasome activity, and treating proteinopathies.
