abstract | The invention relates to a method for preparing an optically pure chiral sulfoxide compound by asymmetrically oxidizing thioether as a precursor of a sulfoxide compound. The method comprises the following steps of: dissolving a D(L)-tartaric acid monoester monoamide compound and isopropyl titanate into an organic solvent; adding purified water equal to the dosage of a catalyst, heating the purified water to 40-60 DEG C, and reacting at the temperature for 0.5-1.5hours; adding the thioether as the precursor, stirring and dissolving, and then, cooling to 5-15 DEG C; adding organic amine, and stirring for 5-15minutes; dropwise adding a peroxide oxidant, and then reacting at the temperature of 5-15 DEG C for 5-7hours; extracting by using 10-15wt% of ammonia water after the reaction is ended, merging water phases and adjusting the pH value to 7-8; and then, extracting by using ethyl acetate, merging organic phases, and carrying out vacuum concentration to obtain the optically pure chiral sulfoxide compound. After the reaction is ended, a target product with the e.e value of 99.5% can be obtained through simple extraction and concentration, and the yield of the target product is up to 70% or more. |
