abstract | The invention discloses a metagenome chip for screening novel genes in an acidic mineral leaching system and a screening method thereof. The method comprises the following steps of: constructing a gene chip which consists of 7766 fosmid plasmids containing microflora DNA (Deoxyribonucleic Acid) large fragments in the mineral leaching system; and screening functional genes of microorganisms which are not separated, purified and cultured through hybridization of the chip with PCR (Polymerase Chain Reaction) products of target genes to be screened. Compared with conventional method based on function expression to screen genes, the method disclosed by the invention greatly improves the utilization ratio of the clone library. Meanwhile, the novel method further provides a feasible path for researching heredity and metabolism of microorganisms which are not separated, purified and cultured. |
