abstract The invention discloses a separation and purification method of tobacco chlorogenic acid. The separation and purification method comprises the steps of: extracting a tobacco chlorogenic acid primary extraction liquid; adding 25-100 mg of active carbon in each milliliter of the tobacco chlorogenic acid primary extraction liquid; stirring and absorbing 30 min at a room temperature; washing residual impurities which are unabsorbed by the surface of a material with water; then eluting the chlorogenic acid by using a 60% ethanol or acetone solution at 50 DEG C; condensing an eluant and pouring the condensed eluant on macroporous resin for absorption, washing, elution; and drying so as to obtain the chlorogenic acid with the purity of the liquid chromatogram greater than 60%; and further refining to obtain high-purity chlorogenic acid. As the active carbon is innovatively utilized to absorb the tobacco chlorogenic acid, the separation and purification method of the tobacco chlorogenic acid obtains the absorption rate of more than 99% and the resolution rate of greater than 95% and low cost, and is suitable for large-scale production; as the active carbon and the macroporous resin are used together, the separation and purification method of the tobacco chlorogenic acid has low loss of the chlorogenic acid and high recovery rate, is easy to operate and saves solvents; and as the absorption difference of the active carbon and the macroporous resin is utilized, the separation and purification method of the tobacco chlorogenic acid can separate impurities with different properties and obtains higher production purity.
