abstract The invention provides a reverse micelle method for separation and extraction of chub protamine. The method is characterized by comprising the following steps: preparing a chub protamine crude extract; conducting microfiltration and ultrafiltration, adjusting pH value and ionic strength of a filtrate; and preparing a reverse micelle solution and a reextraction aqueous solution. In the extraction and reextraction steps, through improvement of proportion of a reverse micelle system, ionic species, ionic strength and pH value of an extraction aqueous phase, temperature of reextraction and additional substances in the extraction aqueous phase in the reverse micelle method, purification efficiency of the reverse micelle method is improved, so that recovery rate of chub protamine activity is higher than 98.5%, and the purification factor reaches 3. The method overcomes defects in an existing reverse micelle technology for extraction of protamine, such as low active recovery rate and low purification factor.
