abstract | The invention discloses a vitrified cryopreservation and thawing recovery method for bursaphelenchus xylophilus. The method comprises the following steps of: (1) mixing bursaphelenchus xylophilus suspension and a cryoprotectant uniformly to obtain a mixture, and balancing the mixture; (2) performing cryopreservation on the balanced mixture in liquid nitrogen; and (3) thawing and recovering, wherein the cryoprotectant consists of dimethyl sulfoxide and yolk. Each process parameter in the vitrified cryopreservation method is optimized, and the category and the concentration of the cryoprotectant optimal for the vitrified cryopreservation of the bursaphelenchus xylophilus and an optimal treatment mode comprising pretreatment, balancing and thawing recovery are screened. By adopting the method for cryopreservation of the bursaphelenchus xylophilus, the frozen bursaphelenchus xylophilus is high in survival rate and reproductive rate; and the method can be applied to cryopreservation of different strains of the bursaphelenchus xylophilus. |
