titleOfInvention Medicament screening method, medicament used for promoting crosslink of extracellular matrix and application of medicament
abstract The invention discloses a medicament screening method, a medicament used for promoting crosslink of extracellular matrix and an application of the medicament. The medicament screening method is used for screening substances having a promotion effect on expression of an LOXL1 gene, and comprises the following steps: A, constructing a second-generation lentivirus vector using a human LOXL1 gene promoter to control expression of ZsGreen; B, infecting human fibroblasts with the second-generation lentivirus vector to construct the human fibroblasts integrated with PLOXL1-ZsGreen elements; and C, screening the medicament, comprising inoculating the human fibroblasts integrated with the PLOXL1-ZsGreen elements into medium, adding to-be-measured substances into the medium of the human fibroblasts, culturing, and determining a promoting effect of the to-be-measured substances on the expression of the LOXL1 gene through detecting whether green fluorescence intensity increases or not. The method provided by the invention is simple to operate, and wide in applicability.
