abstract | The invention provides a synthesis method of 7-alpha-methoxy-3-deacetylcephalothin benzathine. The method comprises the following steps of: (a) performing 7-methoxy substitution reaction between the cephalotin acid and the sodium methylate and halogenated agent in an organic solvent at (-100)-(-60) DEG C, and hydrolyzing the obtained 7-methoxy substitution product to obtain 7-alpha-methoxy-cephalotin acid; (b) transforming the 7-alpha-methoxy-cephalotin acid into sodium salt or ammonium salt in an aqueous solution; (c) performing enzymolysis reaction between the sodium salt or ammonium salt and the deacetylation curing enzyme in an aqueous solution at 10-40 DEG C to obtain a 3-deacetylation product; and (d) enabling the 3-deacetylation product to react with N,N-dibenzyldiaminediacetate in an aqueous solution to obtain the 7-alpha-methoxy-3-deacetylcephalothin benzathine. The method provided by the invention is milder in reaction conditions, simple and convenient to operate, environment-friendly and suitable for industrial production. |
