abstract | The invention discloses a method for constructing high-yield chitinase bacillus thuringiensis through gene knockout, belonging to the technical field of bioengineering. The method comprises the steps of: firstly, with bacillus thuringiensis ATCC35646 (American Type Culture Collection) as an original strain and a temperature sensitive carrier pKSV7 as a framework, constructing a gene knockout carrier pKSV-ued; and then based on a homologous recombination principle, through a method for knocking a yvoA gene out of a strain genome of the bacillus thuringiensis ATCC35646 through a double exchange process, constructing a high-yield chitinase strain CJ212. Through enzymatic determination of chitinase, the capacity of producing the chitinase by the improved strain is greatly improved. |
