abstract | The invention discloses a separation and purification method of raffinose. The separation and purification method of the raffinose includes: 1) filling a gel chromatography column with gel serving as fillers by a wet process; 2) preparing raw material liquid from raffinose concentrated solution, and introducing raw material liquid into the gel chromatography column, wherein the raffinose concentrated solution is obtained by extracting to obtain raffinose extracting solution from degreased cottonseed meal by water or alcohol water solution and subjecting the raffinose extracting solution to activated carbon decoloration and concentration; and 3) eluting with deionized water or ethanol water as eluant, collecting eluant containing the raffinose, and obtaining purified raffinose after rotary evaporation. Gel chromatography is used for separating the raffinose from protein and the like, and the separation and purification method of the raffinose is simple in technological process, simple and convenient to operate, high in separation efficiency and capable of achieving reusability of the fillers and has wide industrialized application prospect. |
